This is a fantastic initiative to train the next generation of pain researchers! Having completed my PhD investigating the psychobiological mechanisms of chronic pain, including the neural correlates of emotional-motivational pain processing through pharmacological fMRI studies, I know firsthand how crucial specialized training in pain neuroscience is. Our recent publication highlights how estrogen signaling impacts metabolism, inflammatory response, and survival pathways in Jurkat E6.1 T-cell leukemia cells.
💡 Key Findings:
Metabolic Shift from Glycolysis to Oxidative phosphorylation:
🟢 17-beta estradiol treatment caused Jurkat E6.1 T-cell leukemia cells to switch from glycolysis to oxidative phosphorylation (OXPHOS), enhancing mitochondrial energy production.
This shift was driven by reduced hexokinase activity (glycolytic enzyme) and increased pyruvate kinase and citrate synthase activity (OXPHOS enzymes). While cancer cells typically rely on the faster but less efficient glycolysis for energy, estrogen treatment pushed them toward the more efficient OXPHOS pathway, which takes place in the mitochondria and generates more ATP.
Inflammatory and Survival Modulation:
🟢 17-beta estradiol increased interleukin-6 expression in Jurkat E6.1 T-cell leukemia cells.
🟢 Estrogen receptor-α (ER-α)-dependent modulation of Nitric Oxide production in Jurkat E6.1 T-cell leukemia cells after treatment with 17-beta estradiol.
🟢 17-beta estradiol increased p-Akt/Total Akt expression in Jurkat E6.1 T-cell leukemia cells.
💡Why This Matters:
These findings suggest that estrogen-driven metabolic reprogramming could be a potential therapeutic target in T-cell leukemia. Understanding the role of estrogen receptors in leukemia cells might provide novel therapeutic strategies for hematological malignancies.
